Here's the best walk to do - its from Borough to the London Eye (west along the Southbank of the Thames)!
Start the morning at Borough market - can try lots of food and the do a really good hot cider. Definitely got some atmosphere.
Then head towards the Southbank past Southwark Cathedral. You walk past the Golden Hind (very old ship from the elizabethan era). Then past the clink (very old famous prison), past vinoplois (wine museum, built on top of an old roman wine warehouse, worth stopping in).
Then your on the South bank. Continue along the south bank and you walk past the globe (shakesperes museum - also worth a quick look round). Keep walking along the river and you get to the millennium bridge with great views of St Pauls (pop over the river for a look round and come back across) and you also are at the Tate Modern (Londons Museum of modern art - lots of free exhibitions). If you keep walking along the river you get to the London Eye. Recommend going on a trip on this as the views of Big Ben and Parliament are amazing.
Sounds like a long walk, but its not at all (could do it in under 2 hours without really stopping anywhere). But its definitely something to do for the day and you get to see lots of things! I always take people along here as its just a bit different and you get to see a few things.
If you do it quickly you can have the afternoon to explore more.

OOh, there is this big clock, but I can't remeber what it is called, but you should go see it.

buckingham palace
big ben
parliament
tower of london
london eye
picadilly
Science Museum, the top couple of floors aren't very intersting, I'd say lowest (I think it's basement, but maybe ground) is the most fun :)
There's also the Tower of London, Buckingham Palace, Natural History Museum, London Eye, Madame Tussaud's, Oxford Circus (not a Circus, just a place with loads of shops)
I highly recommend looking at the web sites of one of the bus tours - and then I recommend doing one when you get there. For example, if you look at the 3 different "routes" on the following web site, you'll pretty much find all the interesting things to do in London (trust me, I lived there for 2.5 years).
http://www.bigbus.co.uk/uk/html/uk_stops.html

When you arrive, take the tour on one of your first days. It will orient you, plus you'll get a great idea of what you'd like to go back and spend more time at. Having done about 8 of these tours in different countries, I suggest you get one with a live speaker as they're much funnier & more entertaining than the recorded versions
I recommend the Jack the Ripper tour. The tours schedules can be found online but they occur nightly and most of the guides are very entertaining. The web-listings will give you more information but the tours leave from around the Tower of London and move, on foot, through certain areas of Whitechapel. The tour really puts a face and feel to the crimes and the time period. Only advice I will give is to not go by yourself as the tour is at night and Whitechapel still isn't the safest place at night.
You must go to Camden market, portabellow row and covent garden. These are all places filled with culture and character!http://www.travelaccommodation.co.uk/toursuk/londonten.htm

If and when you visit London, you will be with the mainstream of tourists flooding the "biggies" such as Big Ben, Piccadilly Circus, and of course Regent Street. However, why not try stepping onto a riverboat afternoon cruise, which is located just behind The London Eye!

I would suggest taking a quick ride on the Eye, then stepping onto the banks and ordering a cheap one-way ticket to Greenwich on a scenic river boat cruise with a tour guide (£6/$10.50), which will last around two hours. From Greenwich, take a bus out to the Bluewater shopping mall built into a quarry - the shops and the scenery are both VERY worth it. Once you're finished, you can return the same way you came.

Also, you will definitely want to escape the hustle and bustle of London city life on a sunny day, so why not trek on the white cliffs of Dover. It's about a two hour trip on a bus.
